"The Kjeldahl Assay
Proteins are boiled with concentrated H2SO4 which oxidises protein to CO2 and NH4+. The ammonia (12-18% of protein weight) then is assayed by titration with alkali or using a colour assay at 440 nm with Nessler's Reagent. This assay is extremely accurate but is quite tedious and is seldom used now. NOTE: The protein solution must be free from nitrogen derived from other sources."
