The hindlimb landing fool-splay test is conducted by holding the rat horizontally a specified height (20 cm) above a table top and dropping the rat. The distance between the hind feet upon landing is measured, generally by putting a drop of ink on the foot pads and dropping the rat onto a piece of paper. The hindlimb landing foot-splay test is popular because of its sensitivity to acrylamide neurotoxicity. Considering the popularity of the splay test and its inclusion in the EPA neurotoxicity-testing guidelines (EPA OPPTS 870.6200), surprisingly little is known about this test. The anatomical basis for the test is unknown, and there is no obvious analogue used by veterinary or human neurologists. It is possible to hypothesize about the neurological basis for the test. The movement of the rat through space likely stimulates vestibular, tactile (vibrissae and other hairs), and visual receptors. And extension of the limbs is an intuitively reasonable postural response in anticipation of landing. However, the rationale for using the width of the response as in index of function is not at all clear. And while an increase in width has been reported for compounds producing a distal axonopathy with neurofilamentous accumulations, the interpretation of a decrease in width is not at all clear. This interpretation problem is curious, because reductions or loss of function is usually interpretable for tests of neurological function. Although the hindlimb landing foot-splay test is popular, the previously discussed tests of proprioception may be easier to interpret and relate to additional functional and morphological effects.
